Adenine di-Phosphate (ADP) is an important physiological agonist that plays a vital role in normal hemostasis and thrombosis. ... In addition, the P2Y12 receptor is also important for potentiation of platelet activation mediated by other physiological agonists including collagen, von Willebrand and thromboxane A2.

What is ADP in blood clotting?

Adenosine diphosphate (ADP) released from platelet dense granules triggers the binding of fibrinogen to platelet receptor GPIIb-IIIa, resulting in the formation of fibrinogen bridges that link platelets into a loose aggregate.

What is the function of ADP in platelets?

ADP not only causes primary aggregation of platelets but is also responsible for the secondary aggregation induced by ADP and other agonists. ADP also induces platelet shape change, secretion from storage granules, influx and intracellular mobilization of Ca2+, and inhibition of stimulated adenylyl cyclase activity.

Why are ADP and thromboxane important to the formation of a platelet plug?

Platelet Activation

The extra ADP and VWF is especially important because it causes nearby platelets to adhere and activate, as well as release more ADP, VWF, and other chemicals. ... Thromboxane is an arachidonic acid derivative (similar to prostaglandins) that activates other platelets and maintains vasoconstriction.

How does ADP cause platelet aggregation?

Acting through cell surface receptors, ADP activates platelets resulting in shape change, aggregation, thromboxane A₂ production, and release of granule contents.

Platelet Adhesion and Aggregation

What triggers platelet aggregation?

Substances such as collagen, ristocetin, arachidonic acid, adenosine 5′-diphosphate (ADP), epinephrine, and thrombin can stimulate platelets and hence induce aggregation. Response to these aggregating agents (known as agonists) provides a diagnostic pattern for different disorders of platelet function.

Why is ADP important?

ADP stands for adenosine diphosphate, and it's not only one of the most important molecules in the body, it's also one of the most numerous. ADP is an ingredient for DNA, it's essential for muscle contraction and it even helps initiate healing when a blood vessel is breached.

What are the 4 steps of hemostasis?

The mechanism of hemostasis can divide into four stages. 1) Constriction of the blood vessel. 2) Formation of a temporary “platelet plug." 3) Activation of the coagulation cascade. 4) Formation of “fibrin plug” or the final clot.

What are the 5 steps of hemostasis?

• Vessel Spasm. ...
• Formation of Platelet Plug. ...
• Blood Coagulation. ...
• Clot Retraction. ...
• Clot Dissolution (Lysis)

What is the role of thromboxane?

Thromboxane is a potent vasoconstrictor and stimulus for platelet aggregation and the reduced vasoconstriction and platelet aggregation that occur may be significant in patients with bleeding tendencies or may complicate surgical procedures.

Is ADP released by platelets?

ADP is actively secreted from platelet dense granules but is also passively released from damaged erythrocytes and endothelial cells. Most platelet agonists, including ADP, activate platelets via cell surface receptors coupled to heterotrimeric GTP-binding proteins or G proteins.

How common is hit?

Patient type: Surgical patients are at greater risk of HIT then medical patients (RR 3.2, 95% CI 2.0-5.4). HIT is uncommon in medical and surgical intensive care units; one study found an overall incidence of 0.4% in this population. HIT is also very rare among pregnant women receiving heparin.

Why do platelets need serotonin?

Serotonin is transported by platelets and released upon activation. This induces constriction of injured blood vessels and enhances platelet aggregation to minimize blood loss. Consequently, serotonin receptor antagonists have been tested for their anti-ischemic potency in atherothrombotic disease.

What are the 3 stages of blood clotting?

Hemostasis includes three steps that occur in a rapid sequence: (1) vascular spasm, or vasoconstriction, a brief and intense contraction of blood vessels; (2) formation of a platelet plug; and (3) blood clotting or coagulation, which reinforces the platelet plug with fibrin mesh that acts as a glue to hold the clot ...

What are the 12 clotting factors?

• Factor I - fibrinogen.
• Factor II - prothrombin.
• Factor III - tissue thromboplastin (tissue factor)
• Factor IV - ionized calcium ( Ca++ )
• Factor V - labile factor or proaccelerin.
• Factor VI - unassigned.
• Factor VII - stable factor or proconvertin.

What is the role of thromboplastin in blood clotting?

Thromboplastin is a mixture of both phospholipids and a posh enzyme that's found in the brain, lung, and other tissues and particularly in blood platelets. Its main function is to convert prothrombin to thrombin within the clotting of blood.

What are five factors that affect hemostasis?

It is affected by the characteristics of blood vessel walls, platelets, the fibrinolytic system, and the coagulation pathway, which are all intimately related ( Figure 1). All these factors function normally to produce an equilibrium between antithrombotic and prothrombotic factors.

What is the cause of hemostasis?

Hemostasis occurs when blood is present outside of the body or blood vessels. It is the innate response for the body to stop bleeding and loss of blood. During hemostasis three steps occur in a rapid sequence. Vascular spasm is the first response as the blood vessels constrict to allow less blood to be lost.

What are the two major disorders of hemostasis?

The most common inherited diseases are von Willebrand disease (primary hemostasis), which is the most common inherited disorder of hemostasis, and hemophilia A (factor VIII deficiency, secondary hemostasis).

What is the process of blood clotting called?

Blood clotting, or coagulation, is an important process that prevents excessive bleeding when a blood vessel is injured. Platelets (a type of blood cell) and proteins in your plasma (the liquid part of blood) work together to stop the bleeding by forming a clot over the injury.

How do platelets heal wounds?

Platelets help prevent blood loss at sites of vascular injury. To do this, they adhere, aggregate and form a procoagulant surface favoring thrombin generation and fibrin formation.

What is normal hemostasis?

Hemostasis is the physiologic mechanism that stems bleeding after injury to the vasculature. Normal hemostasis depends on both cellular components and soluble plasma proteins. Circulating platelets adhere and aggregate at sites of blood vessel injury.

What is the main function of ATP and ADP?

Think of it as the “energy currency” of the cell. If a cell needs to spend energy to accomplish a task, the ATP molecule splits off one of its three phosphates, becoming ADP (Adenosine di-phosphate) + phosphate. The energy holding that phosphate molecule is now released and available to do work for the cell.

Does ADP have energy?

Thus, ATP is the higher energy form (the recharged battery) while ADP is the lower energy form (the used battery). When the terminal (third) phosphate is cut loose, ATP becomes ADP (Adenosine diphosphate; di= two), and the stored energy is released for some biological process to utilize.
